پاسخ های فیزیولوژیکی-بیوشیمیایی بذور زوال یافته بادام زمینی به پیش‌تیمار پراکسیدهیدروژن در جریان تنش خشکی

چکیده

زوال بذر یکی از عوامل مهم کاهنده کیفیت فیزیولوژیکی بذر است که در شرایط نامساعد محیطی خسارت آن شدت می‌یابد. اثر پراکسید هیدروژن در بهبود وضعیت فیزیولوژیکی و فعالیت آنزیم های آنتی اکسیدان بذور زوال یافته بادام زمینی (Arachis hypogaea L.) تحت تنش خشکی طی آزمایشی به‌صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار بررسی شد. پیش‌تیمار پراکسید هیدروژن در غلظت‌های صفر (هیدروپرایمینگ)، 50، 100 و 150 میکرو-مولار و سطوح خشکی صفر، 4/0-، 6/0- و 8/0- مگاپاسکال مورد ارزیابی قرار گرفت. نتایج نشان داد که پیش‌تیمار بذور با پراکسید هیدروژن در سطوح مختلف خشکی موجب افزایش درصد جوانه زنی، سرعت جوانه‌زنی، بنیه بذر، طول ساقه‌چه، طول ریشه‌چه، کربوهیدرات‌های محلول، پروتئین‌های محلول و فعالیت آنزیم های کاتالاز، سوپراکسید دیسموتاز و آسکوربات پراکسیداز و کاهش متوسط زمان جوانه‌زنی، هدایت الکتریکی و محتوای مالون دی‌آلدهید شد. در پتانسیل 8/0- مگاپاسکال، پیش‌تیمار بذور با غلظت‌های 50 و 100میکرومولار پراکسید هیدروژن سبب افزایش سرعت جوانه زنی، شاخص بنیه بذر، کربوهیدرات‌ها و پروتئین‌های محلول به‌ترتیب به‌میزان 89/27، 24/93، 42/71، 12/46 و 06/27، 73/76 ، 26/30 ، 04/51 درصد و همچنین افزایش فعالیت آنزیم‌های کاتالاز، سوپراکسید دیسموتاز و آسکوربات پراکسیداز به‌ترتیب به میزان 37/26، 79/07،8/39 و 43/15، 28/5 ، 51/22 درصد شد. بنابراین استفاده از 50 میکرومولار پراکسیدهیدروژن جهت کاهش اثرات منفی ناشی از زوال در تنش خشکی قابل توصیه است.
